Abstract
Visualizing the synaptic connectivity of identified neuronal populations is an essential part of defining neuronal circuits in the central nervous system. In our laboratory, we have optimized several histological approaches that are compatible with confocal and transmission electron microscopy (TEM) to study the anatomical relationships between sensory afferents, interneurons, and projection neurons in the spinal dorsal horn. The aims of this chapter are to provide detailed descriptions of some of these protocols, to highlight both advantages and potential difficulties that may be encountered when using these approaches, and to reference several studies where these have been applied to study spinal circuits in normal and chronic pain states.
