Abstract

Thirteen viable and frozen-thawed Shami cows embryos obtained by super-ovulation and artificial insemination technique in order to determine the sex of embryos using Polymerase Chain Reaction (PCR). <br/>Biopsies (1-2 cells) of embryos at the morula or blastocyst stage were taken using a micromanipulation system under an inverted microscope. Blood samples from male and female Shami cows were taken as a control. 16 primers were used to amplify DNA fragments using PCR to determine the possibility of Shami embryos sexing. <br/>Results indicated, for the first time that four out of 16 primers (P1sex, P2sex, P1Aut, P2Aut) were determined to help successfully in sexing embryos of Shami cattle by showing bands with a size of 141bp distinguishing male embryos. It was concluded that aspiration of 1-2 blastomere(s) from embryos at morula or blastocyst stage was enough to determine the sex of embryo using PCR which can be used as a useful tool in splitting and transferring desired sex embryos in genetic improvement program.
